I wouldn't find it special in terms of where is this from, if it hadn't the crowd applause in the end. Makes me wonder, what show was it? Was it filmed? Is there any picture of Amy there?
I would hope it is Union Chapel, because it has a presenter as well, a crowd, it is November 2006, but it's not. There is an original file uploaded on Soundcloud by "BBC Big Band", and it is claimed to be: "Recorded live in concert at The Stables Wavendon, November 2006"

In the original file, the end has some more Amy speaking and saying actually " Thank you very much, ok , I'm gonna try..." SO she did more that day,

This has Jonathan Ross speaking in the end, while in this topic's version, Amy herself is speaking in the end.
